The pair built on their inspirational day one performance as play resumed on Saturday, ultimately adding 271 for the second wicket as Cavaliers piled on a ridiculous 6-424 declared at Jack Brabham 2.
While Isaacs was eventually dismissed for 153 after notching his ton last weekend Niederle resumed the day unbeaten on 97 and passed triple figures before being uprooted for 146.
Benny Piper rammed home the maroons' advantage with a quick-fire 48 before the side's declaration came with an unassailable lead of 283 on the board, and a search for maximum points looming.
They almost scored that outright result, but in the end CYMS avoided it and finished 7-189 when stumps was called.
Kinross did pick up an outright victory over Centrals in the other Second Grade fixture, however scorecards were unavailable at time of publication.
Spring Hill remain undefeated and on top of the Third Grade standings after easily accounting for Cavaliers on Saturday, the Wood Ducks successfully chasing the maroons' 107 with nine wickets and 27 overs to spare.
The Gladstone (5-132) upset Centrals Wanderers (130) on Saturday afternoon and Centrals White (175) fell agonisingly short of their first win of the summer against CYMS Kellys (3-185).
CYMS Bourkes (9-219) picked up a thrilling win over Orange City (6-217) though thanks to a fighting, 49-run, 10th wicket partnership between Anand Mohan (33 not out) and Githin Francis (23 not out).
They came together at 9-170 after Abhilash Kaithamangalathu had brought the green and golds back into the game, he came to the crease at 7-60 before belting 84, and guided their side home in the final over.
Their effort defied Lawson Hamling's excellent knock of 98 and Gus Cornish's crucial, wicket-taking spell of 3-57.
Kinross Blue returned to the winners' circle in Centenary Cup, knocking off CYMS by 22 runs.
The students finished with 3-134 from their 35 overs and while Jeron Sijo (64) was excellent in the chase, he didn't much support as CYMS rolled over for 112 in reply.
Will Englund was a big reason for that though, Kinross' strike bowler taking excellent figures of 4-17 to lead his side to victory.
