Perry (2-42) began Australia's assault with the ball against their hosts, when she dismissed Danni Wyatt with the help of wicket-keeper Alyssa Healy soon after England had resumed their innings 3-49.
Not long after that, England were 8-89.
Maddy Villiers' half-century and 24 runs from Ellie Threlkeld helped take the home side's score past 150, before leg-spinner Georgia Wareham (2-31) came on to finish off the tail, adding to the work done by Tayla Vlaeminck (4-31), Perry, Sophie Molineux (1-11) and Ashleigh Gardner (1-14).
Australia had a Perry's (101 not out) century to thank, along with a solid contribution by Jess Jonassen (44), for correcting a poor start to their second innings which had seen the visitors stumble to 4-66.
Australia will begin day three with the score 6-205, and with Perry and Sophie Molineux at the crease.
