The clinic opened the same day as another positive coronavirus case from Orange was confirmed.
Orange Practice manager Ann Carter said it would take up to two days for people to receive their test results.
She urged people to make a booking before attending.
Ms Carter said several people arrived without notice and had to wait.
"We didn't turn anyone away but they had to wait. People just walked across from the hospital. We can't have them waiting around here," she said.
She said the intended process was for people who had booked to wait in their cars at the site outside the Bloomfield Medical Centre.
Ms Carter said they would then be called on their mobile phone to come to the clinic buildings.
Several portable buildings have been set up at the venue for the testing.
Western NSW Primary Health Network CEO Andrew Harvey said the clinic had received an "immensely positive" response from residents.

"To receive such a positive uptake of the service from local residents is exceptional, and shows their determination in really being part of the regional response to COVID-19," he said.
Orange mayor Reg Kidd said the clinic had made a good start. "Anything that streamlines the process of testing during this time is a good thing," he said.
Anyone with mild and moderate COVID-19-like symptoms is urged to book in to have a test at the doctor-led clinic instead of going to Orange hospital.
It's open from 8am-noon and 1pm-5pm Monday to Friday. Book by phoning the Bloomfield Medical Group 5335 6666 or at www.hotdoc.com.au and search for Orange Respiratory Clinic.
