The clothing store, now in Anson Street, was the first shop to open in the old Myer space and occupies the site of the former kitchenware section.
Store manager Kylie Hall said everything in the store was new, the fit out took six weeks, and the Summer Street store would remain open for at least another week.
"We've been at our store for 41 years and 11 months and we were needing an upgrade and we were really excited at the opportunity to join the city centre," she said.
She said being located near the Anson Street crossing would make the store more of a focal point and would also attract more foot traffic.
Orange City Centre centre manager Nicole Chatman said she was excited to see Best and Less open as the first store in the new development and other businesses behind the facade of the former Dalton Brothers building were expected to open in mid-November.
"Best and Less was always going to come on earlier because it is a little bit more separate from the rest of the development," she said.
"I think having a mini major in the centre attracts a lot of traffic into the centre.
"It is really exciting there were so many people here this morning."
Shoppers Chloe and Julie Dittmar were among the first to get a look at the new store and already regular Best and Less shoppers they were impressed with the new store.
"[We came] to see what the new store looked like, it's amazing," Julie Dittmar said.
"It's the best shop in Orange, it's so cheap, it's got good quality clothes."
