The event will again support Little Wings, an organisation which helps sick children from rural areas and their families travel to and from medical treatment in bigger centres.
Tractor Trek president Nick Clancey said the move to centre on Dubbo in 2019 was a result of many families supported by Little Wings being based in the city.
"It's our first time through Dubbo ... a lot of little wings recipients live out that way," he said.
He said the charity had already raised $85,000 and while $100,000 is looming there's no fundraising goal.
The route, which has always changed from year to year and was based out of Molong in 2018, willl head out towards Toronga Open Plains Zoo on Friday, with a planned photoshoot with tractors and giraffes before two longer days.
Saturday will take the tractors through Wongarbon, Beni forest and back to Dubbo Showgrounds, with the charity's annual gala dinner, while the next day will see people heading to Guerie, Wongarbon and back to Dubbo.
"We've had a few extra tractors but not a huge amount of support from around Dubbo itself yet," Mr Clancy said.
"It's hard with it being the first time ... hopefully we'll get it on that day."
The 2019 Tractor Trek will be from September 27 to 29 based at Dubbo Showgrounds.
